According to the FDA, two older persons who got Pfizer's respiratory syncytial virus (RSV) vaccination during a clinical trial were later diagnosed with the rare neurological illness Guillain-Barré syndrome.

Briefing documents (pdf) issued on February 24 in advance of this week's meeting of the Vaccines and Related Biological Products Advisory Committee mentioned the two occurrences of the condition and indicated that Pfizer's vaccine poses a danger.

"Given the temporal connection and biological plausibility, FDA concurs with the investigators' assessments that these occurrences were possibly connected to the study vaccine," the FDA said in the documents. "As a result, [Guillain-Barré] is regarded as a significant potential concern."

In a phase 3 experiment including 20,000 vaccination recipients, two patients in their 60s were diagnosed with Guillain-Barré syndrome after receiving the RSV vaccine. There were no cases of the neurological disease in the placebo group.

One of the patients recovered completely after three months, and another showed signs of improvement after six months. According to the briefing documents, the FDA requested that Pfizer do a safety study if the RSV vaccine is authorized in the spring. The advisory group will meet on February 28 to consider RSV vaccines from Pfizer and GSK for adults aged 60 and up.

According to Pfizer's own briefing material (pdf), the cases of Guillain-Barré syndrome have various reasons that are unconnected to the vaccine. Pfizer discovered no safety problems throughout the trial, and the firm claimed that if approved, it will do a safety study on its RSV vaccine. According to FDA briefing documents, Pfizer's RSV vaccine was 85.7 percent effective at preventing serious illness.

What exactly is Guillain-Barre Syndrome?

Guillain-Barré syndrome is a rare neurological illness in which one's own nerves are attacked by the immune system. The earliest signs are weakness and tingling in the hands and feet.
